    This work refers to an interdisciplinary study on the archaeological site of Pyrgi, an Etruscan harbour still under excavation, located on the Lazio’s coast in Santa Severa, in the province of Rome. The site in question is subject to frequent flooding which compromises its accessibility and delays the archaeological excavation operations. The study is based on the combined use of geomatic technologies, meteorological and climatic models, and hydrogeological knowledge of the examined site, to have a global view of the hazard to which it is exposed. Different geomatic techniques at different scales are used in the analysis. Large scale surveys are carried out to define the water networks and to monitor the site using satellite images. On a small scale, drone photogrammetry techniques are used to assess the morphology of the territory and eventual protection from natural hazards present in the site. Using these images, a detailed digital surface model (DSM) has been generated. The objective of the research is to assess the main cause of the floods and the time the water stays in the site and to determine if the floods are periodic phenomena over time or random events. The study was conducted using images captured by Sentinel 2 satellites processed at level 2-A. These images enabled the identification of the flooding periods of the site for the years of monitoring. The study was conducted by comparing the captured images with rainfall data, paying attention to extreme weather phenomena that occurred from 2012 to date. The rainfall data are provided by the National Department of Civil Protection to CNR-ISAC by an agreement between the two institutions. The same images have been compared with the wind data recorded by the anemometer located in the Civitavecchia harbour and the wave height data available from ERA5 reanalysis. Knowledge of the main cause of the floods and a possible periodicity will allow to plan correct conservation of the site through specific protection measures designed according to the hazards to which it is exposed

    Reconstructing Antarctic Holocene climate/environmental changes from ice and marine cores

    Paleotemperature reconstructions from Antarctic ice cores rely mainly on δD and δ18O records, with the main key factors controlling their observed distribution in Antarctic surface snow being related to the condensation temperature of the precipitation and the origin of the moisture. Reconstructions of past sea-surface temperatures (SST) and sea ice cover (SIC) from marine cores at high southern latitudes mainly rely on diatom-based transfer functions. However, quantitative records of SST and SIC are concentrated in the mid-latitudes of the Southern Ocean and only few records exist in the Antarctic coastal areas. Here we present an overview of the Holocene climate records that have been compiled in the framework of the ESF-HOLOCLIP project, as well as a new isotopic record from the TALDICE ice core, recently drilled in a peripheral dome facing the Ross Sea. One of the main goals of HOLOCLIP is to reconstruct Holocene climate/environmental changes from ice and marine cores and integrate these data in model simulations. The main common features recognized in Holocene climate records obtained from ice cores are a warm early Holocene (from about 10 to 11.5 ka BP), a cool period centred at ~8 ka BP and a secondary optimum peaking at ~4 ka BP. The Holocene climate reconstructions obtained from sediment cores demonstrate a warmer early-mid Holocene hypsithermal followed by a cooler neoglacial with an amplitude and timing of the transitions variable regionally around Antarctica. Though there exist some problems in both ice and marine core records (chronologies, temporal resolution, global vs. regional, annual vs. seasonal), such approach is unique to fuel paleoclimate models and to better understand the ocean-ice-atmosphere interactions at high southern latitudes beyond the instrumental period

    Immune-related pan-cancer gene expression signatures of patient survival revealed by NanoString-based analyses.

    The immune system plays a central role in the onset and progression of cancer. A better understanding of transcriptional changes in immune cell-related genes associated with cancer progression, and their significance in disease prognosis, is therefore needed. NanoString-based targeted gene expression profiling has advantages for deployment in a clinical setting over RNA-seq technologies. We analysed NanoString PanCancer Immune Profiling panel gene expression data encompassing 770 genes, and overall survival data, from multiple previous studies covering 10 different cancer types, including solid and blood malignancies, across 515 patients. This analysis revealed an immune gene signature comprising 39 genes that were upregulated in those patients with shorter overall survival; of these 39 genes, three (MAGEC2, SSX1 and ULBP2) were common to both solid and blood malignancies. Most of the genes identified have previously been reported as relevant in one or more cancer types. Using Cibersort, we investigated immune cell levels within individual cancer types and across groups of cancers, as well as in shorter and longer overall survival groups. Patients with shorter survival had a higher proportion of M2 macrophages and γδ T cells. Patients with longer overall survival had a higher proportion of CD8+ T cells, CD4+ T memory cells, NK cells and, unexpectedly, T regulatory cells. Using a transcriptomics platform with certain advantages for deployment in a clinical setting, our multi-cancer meta-analysis of immune gene expression and overall survival data has identified a specific transcriptional profile associated with poor overall survival
